چکیده
Invasive pulmonary aspergillosis (IPA) is a life-threatening form of fungal infection, primarily in immunocompromised patients and associated with significant mortality. Diagnostic procedures are often invasive and/or time consuming existing antifungals can be constrained by dose-limiting toxicity drug interaction. In this study, we modified triacetylfusarinine C (TAFC), the main siderophore produced opportunistic pathogen Aspergillus fumigatus (A. fumigatus), antifungal molecules to perform susceptibility tests molecular imaging. A variation small organic (eflornithine, fludioxonil, thiomersal, fluoroorotic acid (FOA), cyanine 5 (Cy5) activity were coupled diacetylfusarinine (DAFC), resulting “Trojan horse” deliver compounds specifically into A. hyphae major facilitator transporter MirB. Radioactive labeling gallium-68 allowed us vitro characterization (distribution coefficient, stability, uptake assay) as well biodistribution experiments PET/CT imaging an IPA rat infection model. Compounds chelated stable gallium used for tests. [Ga]DAFC-fludioxonil, -FOA, -Cy5 revealed MirB-dependent active growth inhibition at 16 µg/mL after 24 h. Visualization lungs was possible gallium-68-labeled using PET/CT. Heterogeneous patterns immense influence moiety conjugated DAFC. Overall, novel conjugates promising possibility PET combine both therapeutic diagnostic potential theranostic compound caused fumigatus.

Voriconazole is a broad-spectrum antifungal agent that is effective against moulds such as Aspergillus fumigatus. It inhibits the cytochrome P450-dependent 14-a–lanosterol demethylase, preventing the conversion of lanosterol to ergosterol. This results in the accumulation of toxic methylsterols in the fungal wall and the inhibition of fungal growth [1]. Voriconazole is available as an intraveno...

SUMMARY Aspergillosis is an infectious disease caused by genus aspergillos species. Aspergilli are ab undant in the environment, they Jive in soil as saprophyte deriving nutrient, from dead plants and animal matters. The genus aspergillos in characterized by the formation of distinctive conidiophores giving rise to conidias which can spread by air and are able to reside on the body surface spe...

Antifungal prophylaxis for patients at high risk for aspergillosis is appealing due to the high morbidity and mortality of aspergillosis despite antifungal therapy (1–3). While voriconazole is a logical option, reports of breakthrough mucormycosis are a cause for concern (5). To identify alternative prophylactic strategies, we utilized our novel, inhalational model of pulmonary Aspergillus fumi...
متن کاملNewer combination antifungal therapies for invasive aspergillosis.
Optimal therapy for invasive aspergillosis is unknown, and many clinicians have attempted to utilize a combination antifungal approach to improve outcomes. However, while numerous in vitro studies, animal models, and clinical reports suggest the possibility that combination antifungal therapy might offer improved results, there is no definitive accepted strategy. The currently available antifun...
